Identification of anti-oomycotic compounds in leaves of licorice (Glycyrrhiza glabra) using non-targeted metabolite profiling

During the past years, the use of leaf extracts of Licorice (Glycyrrhiza glabra) for plant protection against different oomycetes (downy mildew, Phytophthora spp.) in organic horticulture has been investigated. Previous studies of a small number of licorice genotypes identified three flavonoids as putative anti-oomycotic principles. However, a recent investigation on basis of a larger set of licorice genotypes suggested, that anti-oomycotic activity is not clearly correlated with the level of these three compounds. This project aims at the identification of further anti-oomycotic compounds from Liquorice leaves using a non-targeted analytical approach. Therefore, leaf extracts prepared from a large collection of licorice genotypes will be comprehensively analyzed by UPLC/ESI-QTOFMS. The resulting metabolic profiles will be correlated with bioactivity data obtained from whole leaf extracts. Leaf metabolites showing a positive correlation between their level and overall bioactivity will be characterized by accurate-mass tandem mass spectrometry. For further bioactivity studies, isolation of these compounds is envisaged. The study is carried out in cooperation with Trifolio-M GmbH providing plant material and bioactivity data.
