This project is aimed at functional characterization of terpene synthases (TPS) from carrot. To this end, candidate TPS genes will be cloned and expressed in Escherichia coli and Nicotiana benthaminiana (tobacco). Intact proteins will be purified from E. coli and tested for their enzyme activity in vitro. Furthermore, the influence of these genes on the conversion and formation of individual terpenes or metabolically related derivatives will be investigated in vivo by Agrobacterium-mediated transient expression. Combination of these two approaches allow understanding of the functional diversity of individual TPS genes.
