Purpose of research: Experimental development

Development of an efficient and genotype independent wheat microspore culture for adopted winter wheat breeding material and wheat genome donors The project was performed to improve the efficiency of doubled haploid production in winter wheat. Therefore new media compositions, stress treatments and donor culture conditions were analyzed. Furthermore so-called inducer chemicals and gametocides were tested. Besides the distinguished tissue culture responsiveness of the variety 'Svilena' was evaluated genetically. To this end DH lines between 'Berengar' x 'Svilena' produced by maize wheat technology were repetitively phenotyped and characterized by molecular markers. Using modified growth conditions for donor plants and innovative stress treatments, it was possible to reach promising regeneration rates. On that account it is assumed that there is a well-founded chance for further acceleration of winter wheat microspore culture to an economically competitive technology in comparison to currently existing DH technologies.
