Purpose of research: Basic research

This project aims at (1) exploiting next-generation sequencing technologies to sequence the completely unknown genome of the fungal pathogen Ascochyta rabiei, and at (2) deciphering its completely unknown transcriptome (the entirety of expressed genes). The expected results will dramatically further our understanding of this pathogen and the infection process, that is unknown at present. The AscoSeq project will provide huge sets of DNA and transcript sequence data, that require intensive bioinformatics for deep evaluation. The direct clients will not participate in this process (except selected scientists from ICARDA itself as visitors in Frankfurt). However, the deciphering of the infection process will certainly catalyze research in at least two CGIAR centers (ICARDA, ICRISAT) and various NARs (and beyond), initially focusing on the underlying genes, with the aim to discover the difference between the various Ascochyta pathotypes and, in not-too-far future, to screen whole fungal populations in ICARDA‘s mandate region for avirulence genes and to design new strategies to predict spread of new mutants (characterized by e.g. up-mutations in key virulence genes). These genes are then known by sequence, which allows the design of specific primers and probes for diagnostic TaqManTM assays, that can easily be performed by the clients (virtually in every NARS or national university laboratory).
