Purpose of research: Experimental development

The major goal of the project was the increase of loging resistance and the associated yield potential through the generation of novel allelic diversity in semi-dwarf genes. This principle was demonstrated using the two target genes BRASSINOSTEROID INSENSITIVE 1 and SEMIDWARF 1 that are involved in the regulation of plant height. In the present study, site-specific mutations were induced in wheat by pollination with maize plants carrying cas9 and guide RNA (gRNA) expression units for wheat target genes. While mutations were induced in the wheat target motifs, the transgene-carrying haploid maize genome was lost during early embryogenesis via uniparental genome elimination. After colchicine treatment for whole genome-duplication genetically fixed and fertile mutant plants were generated. Mutations were obtained in six different genetic backgrounds comprising four bread and two durum wheat genotypes. Progenies showed phenotypic variability in plant height.
