Development of methods for cleavage of azo dyes by human skin bacteria in vitro

Preceding to toxicological investigations of the metabolism of the azo dyes (e. g. direct blue 14, disperse orange 3) analytical methods and appropriate methods for the sample preparation (e. g. from cultures of bacteria) shall be developed and validated.
For the quantification of azo dyes and splitting products/metabolites in extracts of textiles and in mixtures from toxicological investigations, e. g. with the in vitro skin model, analytical methods for the mentioned substances were established. In addition to photometric methods for the quantification of azo dyes such as direct blue 14 and disperse orange 3 various HPLC methods were established. A HPLC method for measurement of small concentrations was established in particular for application in the in vitro skin model. For all these methods standard test procedures were compiled and validated.
