Purpose of research: Applied research
Results from the Project ?Detection and analysis of hepatotoxic modes of action for the prediction of carcinogenic effects of chemicals in the subacute toxicity test (repeated dose 28-day oral toxicity study/ OECD TG 407)? financed by BMBF, indicate towards potential biomarkers and pathways relevant to the early detection of chemically induced cancerogenesis. The relevance of such candidates must be further validated by additional semiquantitative methods, like Western Blot. The most citated empirical tumormarker in the literature is the overexpression of GSTP1 in preneoplastic foci, also detected by our analysis. The precise role of GSTP1 as well as additional pathways involved in cancerogenesis is still unclear and should be investigated in detail. Our aim is the performance of such analysis in the hope of identification of class-specific differences between gen- and nongentoxic cancerogens to be possibly implemented into the regulatory hazard assessment process.
