Purpose of research: Applied research

Consumers are exposed to multiple residues of different pesticides via their diet. Since toxicological properties of substances are generally tested for regulatory purposes on an individual basis, there is only limited data available on potential mixture effects. Hence toxicological investigation of combined effects of substances represents a key challenge to current experimental and regulatory toxicology. Consequently, the development of in vitro tools for assessment of mixture effects is of tremendous importance. In the proposed project, potential mixture effects are exemplified upon a group of widely used fungicides: the triazoles. They are known to cause liver toxicity and endocrine disruption by a common mode of action. Therefore this project focuses on the analysis of a set of human cell lines from endocrine-related organs and from liver by parallel collecting and subsequently integrating data from multiple ‘omics’ levels as a basis for systems biology predictions, covering in particular the metabolism. A comparison to existing results from a classical in vivo study, which was recently conducted at the BfR, will give answer, if the used in vitro systems are suitable to predict combination-effects.
