Purpose of research: Basic research
This project aims at the construction of a physical, BAC-based map of the sugar beet genome. Such a map is of central strategic importance for marker assisted breeding, for straight-forward positional cloning of genes, and for the integration of molecular resources that have already been generated by German sugar beet breeders. Map construction will be based on the hybridisation of specific 35mer oligonucleotide probes to high-density BAC filter arrays, a method that has been very successfully applied to BAC map construction for other genomes. Probes will be designed using bioinformatic tools and existing EST sequence data as well as BAC end sequences generated in this project. In addition, probe design will rely on information about repeat sequences present in the sugar beet genome, both existing and newly generated. The combination of expertise in plant genome research, physical mapping, genome structure and sugar beet repeat sequences will ensure that this consortium will reach the final goal of a high-quality physical map of sugar beet.
