Purpose of research: Basic research
It is the aim of this dissertation project to characterize the alterations in insulin synthesis and secretion in PVDRI pigs by in vivo and ex vivo experiments. Because hypocalcemia is a strong factor influencing insulin secretion, it is a prerequisite for this subproject to establish the methods necessary to normalize calcium and phosphorus blood levels in PVDRI pigs. In vivo insulin secretory capacity in normo- and hypocalcemic PVDRI pigs will be determined by oral and parenteral glucose tolerance tests. Using a number of different stimuli, insulin secretion in isolated islets from wild-type and PVDRI pigs will be measured in ex vivo experiments.
