Proof of sensitization in human dendritic cells of the skin in a migration assay and induction of T cell response in in-vitro models without animal experiments

Purpose of research: Applied research
In current research projects the activation of innate and acquired immune system by den-dritic cells is investigated. Nevertheless to date there are no broadly accepted reproducible cell culture models including primary dendritic cells, which react towards sensitizing substances. In this project the two populations of dendritic cells that localize in human skin, Langerhans cells and dermal dendritic cells, shall be isolated from plastic reductions and will be tested for their response towards sensitizers. Therefore these cells will be activated by known contact allergens. Established stimulation models with cell and virus particle-free bacterial and viral ligands such as peptidoglycan, ds RNA and CpG oligodeoxynucleotides will be performed as reference experiments. Responses of dendritic cells shall be measured by functional assays. Because activated dendritic cells acquire the capacity to migrate and to drive naïve lymphocytes into cell division, cell migration assays and detection of the fluorescent dye CFSE by FACS shall be developed in the lab to quantify cell responses towards sensitizing substances.
