Optical probe technique for the assessment of multidrug resistance profiles in 3-dimensional in vitro tumor models

Purpose of research: Applied research
The current research proposal intends to establish a technically elaborated screening routine for multidrug resistance reversing agents. This routine is based on the optical probe technique which allows to monitor fluorescent model substances for drug transport in the depth of tumor tissues. In parallel, reporter gene vectors of different drug resistance transporters based on green and fluorescent protein will be constructed. These reporter gene constructs will be expressed in different cancer cell lines (e.g. small cell lung cancer, prostate carcinoma, mammary carcinoma, hepatoma, melanoma, glioblastoma). After cultivation to multicellular tumor spheroids we will have a tool to analyze by the optical probe technique the expression of multidrug resistance transporters in living tissue. In parallel the optical probe technique allows by the use of fluorescent marker molecules to analyze drug transport kinetics in double fluorescence imaging routines.
