IdeMoDeResBar aims at the isolation of major genes and Quantitative trait loci involved in the resistance against barley yellow mosaic virus disease (BaMMV/BaYMV), leaf rust (Puccinia hordei), net blotch (Pyrenophora teres) and scald (Rhynchosporium commune). Furthermore, it aims at editing two already isolated genes against barley yellow mosaic virus disease. This will on the one hand lead to a deeper understanding of the structure and function of resistance genes and will on the other hand contribute to a more directed use of genetic resources as a prerequisite for breeding resistant cultivars followed by their use in consumer and environmental friendly barley production systems.
The project is structured in four working packages, i.e. (i) fine and physical mapping of targeted resistance loci and identification of candidate genes, (ii) bioinformatic analyses and data integration, (iii) functional analysis of candidate genes and genome engineering of two virus resistance genes against barley yellow mosaic virus disease, and (iv) marker assisted backcrossing and use of novel resistance genes in hybrid breeding. For gene isolation Genotyping by sequencing (GBS) and Renseq will be conducted and genomic resources, e.g. available ExomeCapture data and the barley reference sequence, will be employed. For the functional analysis of candidate genes and the in planta generation of resistance alleles in elite material we will use an established platform of RNA-guided CAS nucleases. Respective resistance genes will be transferred to adapted breeding lines by marker assisted backcrossing and in a next step used for hybrid breeding.
Leaf rust and Barley mild mosaic virus (BaMMV) are important diseases of barley. The aim of this project is fine mapping and isolation of resistant genes RphMBR1012 and rym15. For RphMBR1012 a high resolution map from 4775 F2 plants was constructed using a set of 55 markers. The target interval was delimited to 0.45 Mb containing 29 genes. For rym15 two maps were constructed using 527 F2 plants were constructed and the gene harbouring interval delimited to 140Mb. In a next step, 5,000 F2 plants will be screened and develop a high density and resolution map of rym15 will be constructed.
