Gastric cell lines to substitute native cells of the gastric gland to study acid secretion in special consideration of preclinical studies

Purpose of research: Applied research
Novel approaches to study gastric acid secretion in vitro focus on the substitution of the usage of animal. In vitro models such as isolated gastric glands or gastric exocrine cells such as the gastric parietal cell bear several disadvantages. As these isolates from stomachs of animals are only to culture for several days, the complicated multi-step preparation has often to be repeated which increase the use of animals. Specific challenges arise from the sensitivity of gastric epithelial cells in in vitro primary cell culture. Epithelial cells are integrated in their tissue, mutually communicating with neighbouring cells and connected to an extracellular matrix. As their lives depend on these contacts, they normally die if they lose them. Losing touch to the matrix induces a special type of apoptosis, named anoikis from the Greek for homelessness. One of our aims is to prevent anoikis of epithelial cells and to apply for longevity. As a long-term goal we try to establish a reliable and accepted method of creating long-living cells or immortal cell lines to study mechanisms of gastric acid secretion to reduce the use of animals or provide an alternative.
