Improvement of the in vitro cultivation of black locust (Robinia pseudoacacia L.) by repression of plant-associated bacteria in combination with their characterization

Purpose of research: Applied research
During the in vitro cultivation of black locust (Robinia pseudoacacia L.) plant-associated bacteria were found, which caused a proliferation disturbance during certain steps of micropropagation. To date, the presumably endophytic living bacteria are repressed by penicillin application.
Within the workings for the diploma possibilities are to be proved to avoid the application of antibiotica. Thus, some antibiotically effective essential oils will be examined for their suitability for in vitro culture use and additionally, a heat treatment method (thermotherapy) will be tested.
Furthermore, more particular insights into the bacterial genus spectra are to gain. For that purpose, bacteria from black locust shoots will be isolated and cultivated on appropriate media, selected by randomised picking and subsequently, will be genetically analysed.
