The project SUNRISE aims at broadening the spectrum of crops for oil production in European markets. Within the project fundamentals for genomics-assisted breeding are to be established as the ultimate prerequisite for long-term competitiveness of sunflower. To identify resistance genes against Plasmopara halstedi (Farlow) Berlese & de Toni, the causal agent of downy mildew in sunflower, innovative breeding strategies will be applied.
High-throughput sequencing and genotyping are used for the characterization and cloning of the PlARG resistance locus on linkage group 1. The detailed haplotype analysis of the gene locus will accelerate the marker-assisted transfer of this resistance into elite breeding material.
Working plan
Using a F2 mapping population, the PlARG resistance locus will be fine-mapped on linkage group 1. Recombinant F2 lines will be identified by SSR marker analysis and subsequently phenotyped in F2:3 which enables fine-mapping of the target region. A further genotyping of single nucleotide polymorphisms (SNPs) and their verification will increase the marker density and support a higher genetic resolution.
A targeted sequencing approach based on the mutation mapping / SHOREmap approach will be pursued to obtain sequence information from phenotypic contrasting bulks (resistant and susceptible) as well as from the resistance donor as a reference. By means of identified SNPs between the bulks, a prediction of resistance gene candidates on linkage group 1 is made.
