The new cooperative research project 'VitiFuture' aims to characterize the defense mechanisms of grapevines carrying resistance locus Rpv10 against Plasmopara viticola (downy mildew). This locus originates from an Asian wild species of grape and is complementing resistance sources from American origin (e.g. Rpv3) in breeding for sustainable resistance against the important pathogen. Detailed comparative microscopy will be conducted to investigate cellular defense reactions of grapevines carrying this P. viticola resistance locus. Additional molecular studies are planned. A BAC (bacterial artificial chromosome) library form the Rpv10 carrying grapevine cultivar `Solaris´ was recently constructed. This library has to be screened with locus-linked markers to identify clones from the Rpv10 region. These clones need to be assembled, aligned to each other and sequenced to identify encoded candidate genes. For functional characterization these newly identified candidate genes will be studied in regard to their activity and regulation in response to pathogen attack. This will be achieved by differential gene expression studies after controlled P. viticola inoculation. Furthermore, parallel metabolomic studies will be performed to analyze the formation of specific defense molecules and link them to the activity of candidate genes. All this work aims to improve our understanding of the resistance mechanism encoded by the Rpv10 locus and to delimit its genes to allow its precise identification and combination in cross breeding minimizing the co-transfer of genomic fragments from the wild species resistance donor.
