Purpose of research: Experimental development
The hemibiotrophic soilborne fungus Verticillium longisporum represents one of the important pathogenic fungi in oilseed rape. So far, only minor genetic variation in resistance to the fungus could be identified in oilseed rape germplasm. To develop resistance against this fungus, the VertiRes project followed a strategy to implement our findings from Arabidopsis thaliana into oilseed rape, in which three genes proved to be functional as putative compatibility factors and knock-out of these genes significantly reduced plant susceptibility to the fungus. To identify corresponding oilseed rape mutants, an EMS-mutant population was creened by TILLING as well as by a NGS-based “keypoint” approaches, leading to identification of several mutants for each candidate gene. As demonstrated by infection experiments, a number of mutant lines displayed significant resistance or reduction in their susceptibility to the fungal infection as expressed by significantly less fungal DNA and reduced disease symptoms in plants. These mutants provide a novel genetic resource for breeding for V. longisporum resistance in oilseed rape and unique materials for a deeper understanding of molecular plant-fungus interactions as well.
