Purpose of research: Experimental development
Main objective of the project is the development of a biochip array which allows a multiplex analysis of the pest thrips at any developmental stage (eggs, larvae, prepupae, pupae and adults) including the tospoviruses they transmit. Modul 1 creates the basis for the visual diagnostic LucID3.4 key through thrips collection, DNA extraction and integument preparation. In Modul 2 the ITS1 and ITS2 regions are amplified and digested by restriction enzymes. Data which are efficient for diagnosis are integrated in a data base for online identification. Modul 3 encompasses the sequencing of thrips and virus material and the determination of especially heterogenous nucleotide sequences and the design, testing and optimization of the complementary oligonucleotide sequences. The electronic proto-biochip and the ID software containing the molecular online identification are applicable in diverse practically relevant and research areas for exact, cost-efficient and fast but also for flexible on-site diagnosis of thrips and tospoviruses.
