Analysis of HVX distribution in generative plant tissue using electron microscopy

Seed transmission of HVX has been reported for the hosta cultivar 'Blue Cadet'. Choi et al. (Acta Biologica Hungarica, 2012) localized the virus in the seed embryo but not in endosperm or seed coat. We will investigate seeds from HVX infected N. benthamiana using immune electron microscopy for detection of HVX as well as evaluation of transmission rate in seeds. Furthermore HVX distribution in generative tissue will be analysed in ultra-thin sections of embedded seeds using electron microscopy. The obtained results will allow comparable analysis of HVX distribution in vegetative/generative tissue among the monokotyle host hosta and the dikotyle host tobacco (Nicotiana benthamiana).
The generated data will be essential for our understanding of HVX epidemiology and estimation of risks regarding virus spread. In 2013 HVX has been included within the EPPO alert list.
