Purpose of research: Applied research

The objective of the APHARES project was the identification of markers for resistance genes against Aphanomyces euteiches, one of the main pathogens of the root rot disease complex of peas. By crossing a resistance donor with an elite line a population of 316 F2-plants was developed and a high resolution genetic map was generated by using the GenoPea-microarray. QTL analysis identified 9 resistance loci with LOD-scores above or slighly below the genome-wide statistical significance threshold of 3.8. Of the 15 developed KASP markers a combination of 3 markers was most informative to identify individual plants with increased resistance/tolerance. A validation of this marker combination for a total of 278 plants of an independent F2:3 population was done in two runs. For the second run a correlation between the resistance degree predicted by markers and the identified resistance by phenotyping could be confirmed. Since the correlation is just r = 0.3, phenotyping is still necessary for breeding, but this could be done with a significantly reduced number of plants.
