Purpose of research: Applied research
The aim of the project is the microscopic and molecular characterization of the Malus wild species M. baccata jackii against premature leaf fall (Marssonina coronaria). The pathogen causes chlorotic leaf discoloration and premature leaf fall. As consequence the tree is physiologically weakened. The fungus appears in organic apple production and infests besides conventional apple cultivars also scab resistant cultivars like 'Topaz'. According to the current state of knowledge, the wild species consists of a high field resistance to Marssonina coronaria. In first artificial inoculation trials in the laboratory no infestation was indicated. If the pathogen causes symptoms after artificial inoculation in a greenhouse based approach was not proofed. (1) At first, leaves of M. baccata jackii will be comparative inoculated in the field, greenhouse and laboratory. (2) Secondly, microscopic sections of infested leaf material from M. baccata jackii and the susceptible cultivar Idared will shed light into the spread of the pathogen in resistant and susceptible plant tissue. Moreover the development of reactive oxygen species and the expression of relevant genes after pathogen infestation will be investigated. (3) In the third part of the project, the inheritance of the trait is the focus of the investigation. Plants of a biparental cross population between Idared × M. baccata jackii will be artificial inoculated in the greenhouse. The aim of this approach will elucidate if one or more loci are associated to the resistance. The results contribute to the question if M. baccata jackii is a suitable resistance donor for further breeding purposes. The project contributes to a reduced application of plant protection agents and an ecological, sustainable fruit production.
