Purpose of research: Experimental development

The project aim is to compare the efficiency of various CRISPR/Cas nucleases for targeted mutagenesis in the tomato genome. DNA breaks shall be induced as a prerequisite for targeted genome modifications (gene targeting). Furthermore, a newly developed CRISPR-based system will be applied for transient downregulation of specific genes which are involved in the mutagenic NHEJ-mediated repair pathway. This mechanism referred to as transient repression shall consequently favor the utilization of homologous recombination for highly precise DNA repair and thus allow for an increased efficiency of gene targeting. This technique shall be applied for future breeding of tomato varieties to obtain highly specific and plannable modifications in genes for improved traits like e.g. yield or resistances.
