Development of phenotypic and genotypic tools for improving the breeding of durum resistance to two mosaic viruses

The cereal mosaic virus (SBCMV) and the wheat spindle striated mosaic virus (WSSMV) are two viruses, present in France, which significantly impact the yield of both common and durum wheat. Several resistance genes providing improved resistance to SBCMV or WSSMV have been identified for soft wheat but not in durum wheat. Thus, breeders are currently relatively powerless to select durum wheat varieties resistant to these two viruses.This project therefore aims to offer several solutions to the breeder:- The main objective of the project is to develop a test for resistance to these two viruses under controlled conditions. Once this test has been developed, the breeder will be able to select his material more efficiently, since the expression of the disease is difficult to control in the field. - The project will also improve the location of the chromosomal regions (QTLs) of the 'Dic2' durum wheat line involved in resistance to SBCMV and WSSMV. Kaspar markers linked to these QTLs will be developed to make these QTLs more easily usable in breeding. - Finally, the project proposes to identify molecular markers closely linked to the Sbm2 gene for SBCMV resistance in the 'Meridiano' durum wheat variety using a new high-throughput sequencing approach on NBS LRR (RenSeq) gene families as well as a fine mapping approach. Gene markers will be validated on durum and soft wheat varieties known to have Sbm2. Thanks to these molecular markers linked to Sbm2, the breeder will be able to easily use the Sbm2 gene in breeding and produce new varieties of soft and durum wheat that are more resistant to SBCMV.
