Validation studies of in vitro alternatives to the Draize rabbit eye test have revealed that cytotoxicity test of simple corneal cell cultures are only valid in combination with organ cultured test from animals for the prediction of potential ocular irritancy. The use of a human corneal model may close the gap between human and animal on the one hand, and between simple cell culture and complex tissue on the other hand. The aim of this project is the construction of a 3-dim corneal equivalent from SV40-transformed corneal cell lines. ; ; The immortalized epithelial and endothelial cell lines are established by Araki-Sasaki et al. (1) and Bednarz et al. (2), respectively. To obtain an immortalized human corneal keratocyte strain, primary keratocytes were isolated from eye bank donor material and transfected with an mammalian expression vector containing genes encoding SV40 early region.
