Expression of several genes encoding transcription factors (TFs) of the so-called NAC family have been found to be induced during senescence of leaves in Arabidopsis thaliana and other plants, including crops. Up to date, however, the knowledge about the functional role of NAC TFs with respect to the control of senescence processes is limited. Here we propose to analyse the regulatory functions of NAC TFs in Arabidopsis and barley (Hordeum vulgare). The Arabidopsis work will concentrate on the following aspects: (i) the identification of target genes for NAC TFs known to control senescence; (ii) the identification of upstream signalling elements that control NAC expression; and (iii) the functional analysis of novel NAC TFs ex-hibiting senescence-dependent expression. In barley we will first identify senescence-regulated NAC TF genes, using multi-parallel quantitative RT-PCR-based expression profil-ing. Through co-operation with partners expression of NAC genes will be altered in trans-genic barley plants and the effect on leaf senescence, plant growth and resource allocation will be tested using various biochemical and physiological methods. The combined work on both plant species will provide a frame for transfer of knowledge for use in other crops of ag-ricultural interest.
