Purpose of research: Basic research
As the most frequent cause of chronic osteomyelitis, S. aureus is able to infect osteoblasts and to persist inside a specific intracellular phagosomal compartment. The aim of this project is the characterization of the osteoblast-specific phagosome, which is the key to the development of persistence. The maturation state and the kinetics of formation of the S. aureus-containing phagosome will be studied. In particular, the involvement of autophagy will be defined with regard to the persistence of S. aureus in primary Osteoblasts. Stimulation of Toll-like receptors will be evaluated as a strategy to enhance osteoblast autonomous immunity.
