Purpose of research: Applied research

Alkenylbenzenes are secondary plant metabolites occurring in numerous herbs and spices, and also in some vegetables. Several alkenylbenzenes are genotoxic carcinogens. Particularly high carcinogenic activity was observed with methyleugenol (ME). Moreover, DNA adducts of ME were detected in numerous human tissue specimens. Carcinogenicity and DNA adduct formation by ME require bioactivation to electrophilic metabolites. These metabolites do not only react with DNA, but also with other nucleophilic cellular constituents. In particular, reaction with glutathione was observed. Glutathione conjugates are usually excreted as mercapturic acids (MA) in urine. In this project, we want to develop analytical methods for the detection of MA of alkenylbenzenes, in particular ME, in human urine samples. The MA levels should be used as biomarkers for the actual exposure toward active metabolites of alkenylbenzenes. We strive to use the urinary MA for the identification of exposure sources and individual risk factors.
