HPCT-1E3 hepatocytoma cultures for determination of cytotoxicity, toxicokinetic interactions and cholestatic effects, pre-validation study

Purpose of research: Applied research
Because HPCT-1E3 cells express a lot of hepatocyte specific properties in contrast to other immortal cell lines their suitability as an in vitro model has to been proven, in order to replace in vivo experiments. A positive correlation (without digoxin) of r²= 0.81 between IC50 values on HPCT-1E3 cells and human LD50 values from the MEIC study was observed for twenty cytotoxic drugs. The correlation between LD50 values from human and rat accounts for 0.54 corresponding to the MEIC study. For the IC50 values of toxic substances which are transported by carriers a correlation of 0.975 to data from HepG2 cells was estimated. In conclusion, HPCT-1E3 cells are a good model for studying cytotoxicity and comparable to HepG2 cells in terms of acquisition of pharmacokinetic interactions of drugs which are transported by carriers.
