Development of a cell culture system for stability testing of hepatitis E virus

The number of notified hepatitis E cases in Germany is continuously increasing. Transmission of the causative hepatitis E virus (HEV) through food is suspected. However, data for the stability of the virus in food are still missing. In the project, a cell culture system for determination of the infectivity of HEV will be developed. In the preceding project phase, such a titration system could be developed based on the newly generated sub-cell line A549/D3 and the human HEV isolate 47832. The distinct mechanism of increased virus growth in this system will now be investigated. In parallel, the system will be used for the investigation of the (long term) stability of HEV at different temperatures. Thereafter, the cell culture system will be adapted for the analysis of food samples and the HEV stability in raw and liver sausages will be investigated. At the end of the project, the results shall be used for the assessment of the risk of HEV transmission through distinct food matrices and incorporated into suggestions for handling and treatment of these foods.
